3. Reaction with Calcium, demineralization of bone Osseous tissue is known to contain a high mineral content, which includes a high concentration of calcium (Ca2). For applications involving conventional histology, i.e. microtome sectioning, staining, and visualization via light microscopy, this mineral must be removed prior to bone preparation and sectioning for these applications. For the demineralizing process chelating agents can be used to demineralize bone tissue while preserving the integrity of intercellular structures. One suclh chelating agent, ethylenediaminetetraacetic acid (EDTA), binds to the calcium ions and effectively removes them from the surrounding tissue when introduced into porous bone (EDTA)
